TSKgel® SW mAb Columns
The TSKgel® SW mAb products consist of three specialized columns designed for the separation and analysis of monoclonal antibodies (mAb). While mAb can be analyzed using many different modes of HPLC, size exclusion is best for aggregation, dimer, and fragmentation, making it the best method for heterogeneity studies. The TSKgel® SW mAb columns are:
- TSKgel® SuperSW mAb HR for high resolution analysis of mAb fragments, monomer and dimer
- TSKgel® SuperSW mAb HTP for high throughput analysis of mAb monomer and dimer with reduced internal diameter and length for fast run times (UHPLC compatible)
- TSKgel® UltraSW Aggregate for high resolution analysis of higher order mAb aggregates
The best performance of TSKgel® SW mAb columns can be achieved when using them in chromatography systems with minimized dwell volumes, such as modern UHPLC systems.
The TSKgel® SW mAb columns utilize a unique pore-controlled technology, which produces a shallow calibration curve in the molecular weight region of a typical monoclonal antibody. As shown in the figure, the calibration curve for the TSKgel® SuperSW mAb HR column is similar to that of the TSKgel® G3000SWXL column curve and has a shallower slope than the TSKgel® UltraSW Aggregate column around the molecular weight range of gamma-globulin. This shallow calibration curve produces high resolution separations. The TSKgel® UltraSW Aggregrate calibration curve shows a separation range up to around 2 million Da, which implies better resolution of aggregate/multimer of a mAb.
The TSKgel® SW mAb HTP calibration curve is similar to the TSKgel® SW mAb HR curve, since both columns are packed with the same resin and only differ in column dimensions.
